These results are similar to our earlier findings showing that dietary salt modulates the presystemic disposition of single doses of orally but not intravenously ; administered quinidine.12 In the present study, by using the stable-label approach, we were able to establish that this dietary salt effect is preserved during long-term oral therapy.
MRP-To examine the interaction between SecR and MBP more specifically, in vitro binding studieswere performed with purified wild-t-ype SecB, SecBEy'iK, andSecBL'i5Q proteins. Purified SecB exists as a tetramer 27 ; , and SecBE'i7K and SecBL75Q were indistinguishable from wild-type SecB by gel filtration. The interactionof SecB with purified MBP can be demonstrated directly because binding of purifiedSecB to unfolded MBP blocks the refolding of MBP in uitro 9 ; . The refolding of MBP in the presence and absence of wild-type SecB, SecBE77K, and SecBL75Q was analyzed to determine the effect of wild-t-ype and mutant SecB on the folding of MBP. The refolding of MBP was monitored by measuring the change in the intrinsic fluorescence of tryptophanyl residues as unfolded MBP refolds. Purified MBP was unfolded in 2 M guanidine hydrochloride buffered with 10 mM Hepes. pH 7.6. Refolding wasinitiated by rapid dilutionof the protein into a cuvette containing 10 mM Hepes, pH 7.6, at 4 "C. The fluorescence intensity was measured with respectto time, and the results of a representative experiment are plotted in Fig. 4. As observed previously 9 ; , in the absence of SecB. the fluorescence intensity increases during the refolding of MBP and reaches equilibrium after approximately 30 min. T o measure the effect of SecB on the folding of MBP, purified SecB protein was added to the cuvette prior to the addition of MBP. As shown in Fig. 4, A and R, when wildtype SecB was present in the cuvette at a SecB: MBP molar fluorescence intenratio of 4: l monomer concentrations ; , the sity at equilibrium was lower than that observed in the absence of SecB, indicating that the folding of MBP is blocked. SBP was significantly higher at baseline and was reduced significantly more by indapamide, but this difference was not significant after correcting for baseline pressure. Triglyceride levels were increased significantly more by indapamide, and this remained significant after adjusting for baseline values; however, there was no significant difference between HCTZ and indapamide with respect to their effects on HDL, LDL or the ratio of TC to HDL, which was the main relationship chosen for the study. The only difference in an intention-to-treat analysis was that the triglyceride results were in the opposite direction there was no significant difference in triglyceride levels, but after adjustment for baseline, the indapamide patients had a decrease in triglyceride levels by 0.450.73 mmol L, and the HCTZ patients' triglyceride levels increased by 1.530.45 mmol L; P 0.048.
