As part of a collaboration between the NC3Rs and the European pharmaceutical industry a working group of 15 international companies have been sharing and analysing the value of data from single dose rodent acute toxicity studies. The conclusion from this extensive review is that the information obtained from conventional acute toxicity studies is of little or no value in the pharmaceutical development process. The working group presented its findings to regulators from Japan, Europe and the US. Discussion at the workshop indicated that the recommendations of the working group are likely to be reflected in the new draft ICHM3 guidance. QL-02. ASSESSING CONTENT VALIDITY: DEVELOPMENT OF THE M.D. ANDERSON SYMPTOM INVENTORY--BRAIN TUMOR MODULE MDASI-BT ; Terri S. Armstrong, 1, 2 Tito Mendoza, 1 Marlene Z. Cohen, 1, 2 Marilyn Morrissey, 1 Mark R. Gilbert, 1 and Charles Cleeland1; 1University of Texas M.D. Anderson Cancer Center, Houston, TX; and 2University of Texas-- Houston School of Nursing, Houston, TX; USA Purpose Specific Aims: The aim of this study is to establish the content validity of items to be included in a tool for symptom self-report in the primary brain tumor patient population. Rationale: Establishing content validity is the essential first step in the development of items to be included in a measurement instrument. Content validity is a criterion-referenced assessment that measures how well each item measures its respective objective or content domain in a newly developed tool. Sample: Twenty experts were chosen. Clinical experts met the following guidelines: worked in neuro-oncology for 5 years, had at least one publication related to the care of patients with primary brain tumors, and were considered experts by the brain tumor community. Experts chosen represented many of the disciplines who care for PBT patients, including neuro-oncology, radiation oncology, neurosurgery, psychiatry, neuro-oncology nursing, and neuro-oncology social work. In addition, a patient and caregiver group was also identified to review items for relevance. For analysis of responses, the 20 experts were then divided into 3 groups: patients caregivers; physicians and neuropsychologists; and a nurse social worker group. Methods: Initial steps completed included domain identification and item generation and instrument formation. The completed items were then sent to experts for review, and the content validity index CVI ; was calculated. All experts received the instrument for review in electronic and paper format and were given one month to complete it, and all 100% ; participants returned the questionnaire. The CVI represents the proportion of items on an instrument that achieved a rating of 3 or the experts. A CVI of at least .80 has been suggested to be a good criterion for accepting an item as valid beyond the 0.05 level of significance. The final instrument will consist of items that received a rating of 3 or the experts. Items that did not meet this level of significance were eliminated. Results: The CVI was generated for each item within the three subgroups. Ten items were found relevant by all three groups of experts, CVI .80 ; , 6 additional items by two groups, and 2 additional items were found relevant by the patient group only. Comments regarding rewording and individual scoring of each item and additional items suggested by the reviewers were then reviewed by the research team. Interestingly, one of the most common symptoms found in the literature to be associated with PBT, headaches, was not endorsed by the physician panel. As a result of this process, 18 symptoms were deemed relevant to the PBT population and will be added to the core MDASI for the next phase of instrument development, establishing reliability and validity and monoket.

Background and Objective: The secretoglobulins are a mammalian protein family, no member of which has been indisputably assigned a function. Human secretoglobulin family members include CC10, a protein that correlates with broncho-pulmonary dysplasia and asthma, and MGB and LPGB, proteins overexpressed in breast and prostate cancers. Fel DI is a major cat allergen to which humans react. We have shown that mouse salivary androgen-binding protein ABP ; mediates mate selection behavior, a particularly attractive hypothesis given the evidence for strong positive selection on Abpa. We are pursuing in a broader sense the shared functions of the secretoglobulins with genomic and bioinformatics studies. Experimental Design: We have developed a comprehensive evolutionary picture of the secretoglobulin gene family in the mammalian lines with genomes currently available mouse, rat and human ; by collecting the genes and protein sequences of human and rodent secretoglobulins. We compared the evidence for expression of each protein and the tissues in which each is expressed, and we developed a phylogeny of the expressed proteins which shows their evolutionary relationships. This has allowed us to locate each of the expressed genes within its respective genome. Results: Secretoglobulins separate into five families. In the rat and mouse genomes, these gene families are found in two syntenic clusters. Humans have only the three families that are found in the CC10 cluster; no one has yet identified a protein or gene from the two ABPencoding families. Conclusion: We are using the newly completed mammalian genomes for comparative genomic and proteomic studies to elucidate functions for elusive proteins. Because members of the secretoglobulin family are found in tissues connecting a mammal's outside environment to its internal physiology, they are uniquely poised to be targets of recognition. They function either as the signal of intra-specific recognition ABP ; and or as inter-specific allergens Fel DI ; . Recognition is likely to be secondary to a more basic function, such as coating and protecting surfaces. The absent ABP gene families appear to have been lost in humans, probably concomitantly with the decrease in importance of the accessory olfactory system.

ABSTRACT: The oral fungal microbiota of 30 children with AIDS, of both genders, aged from two to six years, receiving outpatient treatment, was evaluated and compared with that of a control group composed of 30 healthy subjects with matching ages and genders. Virulence factors, such as exoenzyme production, and susceptibility to five antifungal agents using an E-Test kit were evaluated. C. albicans predominated over other species in the AIDS group, showing a higher production of proteinase and phospholipase when compared with that observed in the control group. In this study few clinical manifestations of and low selectivity for C. albicans 23.3% ; were observed in the AIDS group. The enzymatic studies showed that 53.8% of the AIDS strains were strongly positive whereas only 33.3% of the non-AIDS strains were positive. Amphotericin B was the most effective drug among the antifungal agents tested against C. albicans. The frequency, selectivity and level of exoenzyme production by C. albicans suggest a higher pathogenicity in the AIDS children than in the control children. Os estudos enzimticos mostraram que 53, 8% das linhagens no grupo com AIDS foram fortemente positivas, ao passo que apenas 33, 3% o foram no grupo controle. Dentre os agentes antifngicos testados, a anfotericina B foi a droga mais efetiva contra C. albicans. A freqncia, a seletividade e o nvel de produo de exoenzimas pelo microorganismo sugerem a ocorrncia de uma maior patogenicidade nas crianas com AIDS do que nas crianas do grupo controle.
